Un equipo de médicos de una universidad alemana especializado en investigación nefrológica ha desarrollado un nuevo método y kit de ensayo para el análisis de sustancias activas potenciales que se emplea en el tratamiento de enfermedades renales crónicas. El kit utiliza glomérulos aislados (tejido renal intacto) y permite investigar los podocitos en su entorno de tejido natural. El aislamiento se realiza mediante cribado o separación magnética de células. Posteriormente los glomérulos se colocan en recipientes de cultivos celulares con una solución de nutrientes y se almacenan en las condiciones adecuadas. El equipo de médicos busca socios en cualquier región de la EEN que puedan beneficiarse de la tecnología con el fin de establecer acuerdos de licencia o investigación.

Description:
In most cases the cause of chronic kidney disease is an injury of the kidney that results in a glomerular flow rate (GFR) decrease. The GFR indicates the total volume of the primary urine which is required for filtering by the kidney. Instead of the normal flow rate of about 125 ml/min the GFR of a damaged kidney decreases to below 60 ml/min. The starting point of the disease represents the damage and/or deficit of podocytes. Podocytes (visceral epithelial cells) are highly differentiated kidney cells whose injuries or de-differentiation activated signal pathways which lead to cellular mutation and damage within the kidney and which also results in the progression of the disease.

So far, effective ingredients for the therapy of chronic kidney diseases are hardly available. The current search for pharmaceutical active substances is limited to the identification of signalling pathways. Undiscovered pathways remain negligible in the current processes. Alternatively, screenings are performed on podocyte cell cultures. Due to the de-differentiation in the form of cell cultures, the results do not reflect unequivocally the in vivo situation of podocytes.

The innovative medical scientist team of one of the most prestigious Universities of Germany, active in nephrology research, has developed a novel method and test kit for the analysis of potential active substances for kidney disease treatment using a representation of in vivo conditions.

The test kit for screening active substances to podocytes is not based on cell cultures. The invented kit uses the isolated glomeruli (intact kidney tissue) which allow an investigation of podocytes in their natural tissue environment. The isolation is carried out by sieving or magnetic cell separation. Subsequently the glomeruli will be supplied in cell culture vessels with nutrient solution and stored at appropriate conditions. Especially, reporters which allow a continuous monitoring of the cell differentiation are used.

With the assistance of a fluorescent protein a decrease of fluorescence could be documented over the time due to the de-differentiation of podocytes in isolated glomeruli of transgenic mice. It is possible to search for new ingredients that counter-act the loss of the differentiation of podocytes by adding different active substances.

In order to ensure an economical use of their invention and to imcorporate the acquired knowledge into the development of novel applications, the inventors search for partners within the framework of a license or a research cooperation agreement.

Under the preservation of in vivo conditions the method described, enables potential users to examine new active substances intended to treat kidney diseases without the effect of dedifferentiation of podocytes in conventional cell culture environments.
