Description:
A rapid, cost-effective examination of meat adulteration is a very critical issue in food regulatory control. Determining the fraudulent replacement of higher commercial valued meat species by inferior, cheaper or undesirable alternatives is necessary for ethical requirements, specific food allergies, religious affairs, fraud, and malicious marketing practices in addition to safety, economic, religious (Halal testing) and legal concerns.

The technology provider´s rapid multiplex quantitative assay for meat speciation including beef, pork, chicken, sheep, dog and horse as well as meat products and processed meat such as sausage, meatball, etc., is novel and has multiple advantages over currently used meat identification assays.

The Rapid Multiplex Meat Speciation kit developed employs polymerase chain reaction (PCR) amplification in the presence of double stranded DNA binding dye followed by high resolution melting (HRM) technology. Detection of meat species is based on the pre-determined melting point of PCR products. For example, a PCR product which melts at 70 degrees indicates it is from beef and at 74 degrees from pork.

The Rapid Multiplex Meat Speciation kit comprises of the following technologies:

- Primer Design
- Multiplex PCR optimisation
- Optimisation of Melting Point of PCR products so that they can be distinguished from each other
- Innovative method of detection based on the pre-determined melting point of PCR products
Advantages and Innovations:
This method is rapid, sensitive, specific, cost-effective and does not require a probe. Other advantages and innovations of the technology include:

- HRM-based methods provide greater flexibility as there is no requirement for fluorescently-labelled detection probes (such as TaqMan probes), therefore simplifying assay design over traditional real-time PCR methods

- Specific and accurate detection as PCR products can be discriminated according to sequence, length, GC content, or strand complementarity, down to single base pair differences

- Cross-contamination is minimised as HRM is a closed-tube technique. Unlike other genotyping methods such as denaturing gradient gel electrophoresis (DGGE), HRM does not require the use of hazardous reagents such as acrylamide, formamide, and ethidium bromide

- HRM is cost effective as there is no requirement for fluorescently-labelled detection probes
