Description:
The global in-vitro immunodiagnostic market is mainly focused on ELISA (enzyme-linked immunosorbent assay) products and techniques due to their high sensitivity and universal applicability. Yet there are limitations associated with this type of assays, such as lengthy sample preparation and processing times.

The Austrian research organization has developed a novel technology-based solution and realized a prototype for rapid analysis and continuous monitoring. This has been accomplished in collaboration with international partners within the scope of a European project.

The technology is based on adding specially designed functionalized magnetic nanorods (´nanoprobes´) directly to the sample solution (e.g. serum or saliva), and analyze target proteins by optically monitoring the dynamic response of the nanoprobes to applied time-varying magnetic fields. This simple mix -Y- measure approach allows fast and sensitive monitoring of patient parameters without requiring any further manual user interactions.

In addition to quantifying protein concentrations, the technology enables protein size analysis and determination of affinity constants. Multiplex analysis can be accomplished by applying differently functionalized nanoprobes within separate chambers of a microfluidic cartridge.

Currently, the technology has been demonstrated for the detection and analysis of the breast cancer biomarker HER2 (soluble domain) in serum and saliva samples. Here the best results have been achieved at a 1:10 dilution ratio of the sample fluid with the nanoprobe-containing buffer solution.

Potential applications are very broad, including the following examples:

· General point-of-care diagnostics, for example:
- Emergency care patient stratification and monitoring
- Rapid diagnostics at medical and dental offices
- Home based diagnostic for testing of biological samples (e.g. saliva, urine)
· Field testing such as veterinary testing at breeders and farmers
· Protein size analysis
· Binding affinity constant investigations

The organization is seeking life science cooperations who are responsible for product development, marketing and sales worldwide. Collaboration could be in the form of license agreement, technical cooperation or research cooperation.
Advantages and Innovations:
Current immunodiagnostic tests involve expensive detection systems and require lengthy processing time due to shipment of patient samples to central laboratories for measurement. This system has the potential to become the basis for establishing tailored diagnostic tests, which can be used in emergency rooms, ambulances, medical offices or at home.

Compared to state-of-the-art immunodiagnostic tests, the main advantage of the technology is its simplicity and speed. While most established detection methods require multiple reagent binding and washing steps, the technology only involves mixing the sample with a solution of nanoprobes and start the measurement (mix -Y- measure). Thus, possible biases induced by handling errors are greatly reduced, and the analysis time is only determined by the time required for binding of the analyte proteins. In addition, automation of the analysis procedure is much simpler compared to established methods.

The nanoprobe detection system offers the following advantages:
· Only minimal sample preparation required (e.g. dilution)
· Fast total analysis time (< 5 min) due to simple mix -Y- measure technique
· Small sample volumes (20µl currently, 1µl in development)
· Good sensitivity compatible to state-of-the-art techniques
· Easy to integrate and simple instrumentation
